Light and Shadow
No matter what medium used to create, you'll need to think about the direction of light sources around shapes, material type, and how light might reflect off of nearby materials. Mood begins with color selection, so if you find yourself changing the shadows and lighting a lot and don't feel like that helps accomplish the mood you're looking for, you may want to step back and try out new base colors, then try again with light and shadow.
